I’m planning to make my own non-commercial, non-profit, independent animated short film then I’ll upload it on Youtube. I’m just wondering because I liked a soundtrack from a recent film and I want to use about 40 seconds of it as my background music. I’m only making this home made film just as my application for a job as an animator.

You might be able to use 30 seconds or less under the “fair use” clause of the DMCA. 40 seconds might be considered copyright infringement.
